CD Bioparticles Launches PROTAC Linkers for Drug Delivery

CD Bioparticles introduces a series of PROTAC Linkers with various molecular weight and functional groups.

New York, USA – August 31, 2021 – CD Bioparticles, a leading manufacturer and supplier of various drug delivery products and services, now introduces a series of PROTAC Linkers with various molecular weight and functional groups, such as azide, acid, amine, biotin, NHC, tosyl, and fluorine, to accelerate researchers’ PROTAC study.

Proteolysis Targeting Chimeras (PROTACs) are heterobifunctional molecules consisting of two ligands; an “anchor” to bind to an E3 ubiquitin ligase and a “warhead” to bind to a protein of interest, connected by a chemical linker. CD Bioparticles manufactures and supplies various drug delivery products, including polymeric nanoparticles for R&D and commercialization in a variety of application areas, and now is offering a series of PROTAC Linkers for customers, such as (S,R. S)-AHPC-PEG4-Azide,(S,R. S)-AHPC-PEG2-acid, 4-Fluoro-thalidomide, D-Biotin-PEG6-Thalidomide, Pomalidomide-PEG5-Azide, Thalidomide-O-PEG5-Tosyl, Thalidomide-O-PEG6-NHS ester, and VHL Ligand 1 (hydrochloride).

For the (S,R. S)-AHPC-PEG4-Azide, it is a click chemistry PROTAC linker that incorporates an E3 ligase ligand with a PEG4 arm to empower PROTAC research & discovery. The hydrophilic PEG spacer increases the solubility of a compound in aqueous media. Azide enables click chemistry with ALkyne or DBCO, BCN molecule.

For D-Biotin-PEG6-Thalidomide, it is a cereblon affinity probe for PROTAC and targeted protein degradation research. It can be used in cereblon (CRBN) binding and displacement assays for protein degrader validation experiments, CRBN ligand discovery, competition analyses, or where enrichment of CRBN complexes is required. The hydrophilic PEG spacer increases the solubility of a compound in aqueous media.

Other examples, such as 4-Fluoro-thalidomide, the Thalidomide-based Cereblon ligand used in the recruitment of CRBN protein. It can be connected to the ligand for IRAK4 protein by a linker to form PROTAC IRAK4 degrader-1; Pomalidomide-PEG5-Azide, a PROTAC linker that incorporates the Pomalidomide based cereblon ligand and 4-unit PEG linker used in PROTAC technology. Azide enables click chemistry with Alkyne, DBCO or BCN molecules; and the Thalidomide-O-PEG4-Propargyl, a synthesized compound that incorporates the Thalidomide based cereblon ligand and 4-unit PEG linker used in PROTAC technology. Propargyl is reactive with azide molecule in the presence of Cu catalyst.
